She was born Myrna Adele Williams on August 2, 1905, in Helena. Her father was a rancher and a banker. He was also the youngest man elected to the Montana state legislature. Her mother had studied music in Chicago. After her father died during the 1918 flu pandemic, her mother moved the family to California. Loy grew up studying dance in Los Angeles and attended Venice High School.

During her school years, Loy contributed to a lasting piece of art.

She posed as the model for a sculpture titled "Inspiration." This figure was part of a larger group called the Fountain of Education. The sculpture was created by teacher Harry Fielding Winebrenner. It was installed in 1923 in front of the school's outdoor pool. The figure represented purity, grace, and youthful vigor. A bronze duplicate of the sculpture stands at the school today.

Loy's professional career began with small roles in the late 1920s.

She initially worked as a dancer in musical prologues at Grauman's Egyptian Theatre. She was discovered by production designer Natacha Rambova. Early in her film career, Loy faced significant typecasting. She was often cast in "exotic" roles. These included characters of Asian or Eurasian descent or "vamp" roles. She played these parts in films like Across the Pacific and The Mask of Fu Manchu. It took many years of hard work to move past these specific character types.

Her career reached a turning point in 1934.
