He belonged to the famous Barrymore theatrical family. His parents were actors Georgiana Drew Barrymore and Maurice Barrymore. He grew up with siblings Ethel and John, who also became famous performers. This family connection meant that theater was always part of his life. However, Barrymore did not initially intend to follow this path. He actually preferred the arts of painting and music over acting.

In 1906, Barrymore and his first wife, Doris Rankin, traveled to Paris.

He sought training as an artist at the Art Students League of New York. During his time in France, he witnessed Louis Bleriot flying across the English Channel in 1909. Despite his efforts, he did not achieve success as a painter. He returned to the United States in 1909 and rejoined the stage. He eventually found his true calling in dramatic and character acting.

Barrymore's career spanned several different mediums and technical eras. He began with stage plays on Broadway, such as The Mummy and the Hummingbird.

He later transitioned into silent films, working with director D. W. Griffith. He appeared in more than 60 silent films directed by Griffith. In 1926, he signed a major contract with Metro-Goldwyn-Mayer (MGM). This move coincided with the arrival of sound films in 1927. His stage training allowed him to excel at delivering dialogue in these new "talking" pictures.

As a director, Barrymore contributed to the evolution of film technology. He directed several films, including the color film The Rogue Song. This film was the first color production for Laurel and Hardy. Notably, Barrymore is credited as the first director to move a microphone on a sound stage. This was a significant technical step during the early, imperfect period of sound cinema. His ability to manage both acting and directing made him a valuable asset to MGM.

Barrymore reached the height of his professional recognition in the early 1930s. In 1932, he won an Academy Award for Best Actor. This honor was for his performance in the film A Free Soul (1931).

He was also considered for a Best Director award in 1930 for Madame X. Throughout the 1930s and 1940s, he became well-known for specific character types. He often played grouchy or elderly men, which became a recognizable professional stereotype.

Many audiences remember him for specific, iconic roles in film and radio.

He played the mean-spirited banker Mr. Potter in the 1946 film It's a Wonderful Life. On the radio, he was famous for his annual performances as Ebenezer Scrooge. He performed this role from 1934 through 1953, missing only two years. He also played Dr. Leonard Gillespie in nine Dr. Kildare films. Because of a broken hip and arthritis, he often performed this role from a wheelchair.

Barrymore's personal life was marked by both family ties and significant loss. He was married twice, first to Doris Rankin and later to Irene Fenwick.

Sadly, neither of his two daughters, Ethel and Mary, survived infancy. This loss caused great strain on his first marriage. Despite these hardships, he maintained strong connections within the Hollywood community. He remained active in his career until his final film appearance in 1953. He passed away on November 15, 1954.
