He became a director for the company in 1847. He even helped the railroad when it had debt. He did this in 1857 and again in 1873. He was also the president of Merchants' Bank. He was a very successful man in Baltimore. He retired from business when he was fifty-two.

During the American Civil War, Hopkins took a stand. He strongly supported Abraham Lincoln and the Union. His summer home was called Clifton. It was a place where Union supporters met. Some people in Maryland did not agree with him. They supported the South instead. Hopkins also had complicated views on slavery. Some researchers found records from 1840 and 1850. These records suggest he may have owned enslaved people. Other scholars say he was an abolitionist. An abolitionist is someone who wants to end slavery.

Hopkins wanted to leave a great legacy. He was a very kind giver of money. In 1870, he wrote a special will. He left $7 million to help his city. This was a huge amount of money back then. His gift helped start Johns Hopkins University. It also helped start a large hospital. He wanted the hospital to help everyone. He even planned an asylum for Black children. His gifts still help people learn and heal today.
