Led by Jan van Rensburg, 58 families settled near Sergoit Hill. They acquired leaseholds and began ploughing the land with ox-drawn ploughs. They grew wheat, maize, and various vegetables, which turned the plateau into a vital agricultural zone. The settlement was originally called "Farm 64" or "Sisibo." This name referred to its distance from the Londiani railhead on the Uganda Railway. A post office was established in 1910, and the name Eldoret was officially adopted in 1912. The arrival of the Uganda Railway in 1924 sparked significant economic growth.

Infrastructure developed steadily throughout the colonial period. In 1928, a piped water supply was brought in from the Sosiani River. By 1933, the East African Power and Lighting Company installed an electricity generation plant. During the 1950s, the town experienced social segregation. The area was divided along Uganda Road, separating Afrikaner settlers from British settlers. This division influenced where people lived, went to school, and socialized. Following Kenya's independence, the city saw massive investment. President Daniel arap Moi oversaw much of this growth. In 1984, Moi University was established, and the Eldoret International Airport was later constructed.
