They lived on farms in Quebec, New York, Illinois, and Missouri. Eventually, the family settled near Wichita, Kansas. Masterson's early life was defined by the rugged environment of the Western frontier. He worked as a buffalo hunter and a civilian scout. He also served as an Indian fighter on the Great Plains.

Masterson's early career involved several intense and dangerous encounters. In July 1872, he and his brother Ed worked for a subcontractor named Raymond Ritter. Ritter failed to pay the brothers their full wages for grading railroad tracks. Masterson spent nearly a year trying to collect this money. On April 15, 1873, he confronted Ritter on a train in Dodge City, Kansas. Masterson forced Ritter to hand over $300. This money was owed to Bat, Ed, and a friend named Theodore Raymond.

On June 27, 1874, Masterson participated in the Battle of Adobe Walls. He was one of 28 hunters defending the outpost against a siege. This battle involved several hundred Comanche, Kiowa, and Cheyenne warriors. The defenders lost only four men during the five-day fight.

In August 1874, Masterson served as a U.S. Army scout under Colonel Nelson Miles. He helped pursue war parties across the Cherokee Strip and into Texas. During this time, his force worked to recover four young sisters. These girls had been captured by Cheyenne Dog Soldiers. The sisters were part of a family attacked near Ellis, Kansas. Masterson's military service continued through various frontier conflicts. In January 1876, he experienced his first major gunfight in Sweetwater, Texas. A soldier named Corporal Melvin A. King attacked Masterson. During the struggle, a woman named Mollie Brennan was killed by a bullet. Masterson was shot in the pelvis during this event. He recovered but began using a cane, which became a famous trademark.

Masterson later became a central figure in the law enforcement of Dodge City.

As the frontier era closed, Masterson transitioned into new professional roles. He moved to Denver, Colorado, in the mid-1880s. There, he became a "sporting man" and a professional gambler. He developed a deep interest in the sport of prizefighting. Masterson attended almost every major title fight in the United States. He moved to New York City in 1902 to pursue journalism. He worked as a reporter and columnist for The Morning Telegraph. His columns covered boxing and various other sports. He also wrote about crime, war, and politics.
