Francisco Cuervo y Valdés established it as a villa in the province of Santa Fe de Nuevo México. The city was named after Francisco Fernández de la Cueva, the 10th Duke of Alburquerque. It functioned as a strategic outpost on El Camino Real. This famous road linked Mexico City to the northern territories of New Spain. The original settlement followed a traditional Spanish villa pattern. It featured a central plaza surrounded by a church, homes, and government buildings. This area is known today as Old Town Albuquerque or "La Placita."

Major changes arrived in the late 19th century. The Atchison, Topeka and Santa Fe Railroad arrived in 1880.
