The history of this region began in ancient times. The area at the Moche River mouth was a center for many cultures. The Cupisnique culture occupied sites like Huaca Prieta as early as 4700 BC. Later, the Moche civilization flourished from about AD 100 to 800. They were known for sophisticated irrigation systems. These canals diverted river water to supply crops. The Moche also created elaborate painted ceramics and gold work. They built monumental structures called huacas.

This was the largest pre-Columbian city built of adobe. At its peak, Chan Chan had an estimated 100,000 people. During the Late Chimu period, about 12,000 artisans lived in the city. These specialists were grouped into the citadel by their craft. Men often worked in metallurgy and heavy agriculture. Women specialized in ceramics and textiles using cotton and wool. The Chimu used bronze coins for trade.

This was done to honor the home city of Francisco Pizarro. The settlement was built among four existing Chimu towns. On November 23, 1537, King Charles V granted the city a formal coat of arms. This made Trujillo the first city in Peru to receive such a royal decree. By 1544, the city had about 1,000 inhabitants. The economy grew through sugar cane and wheat cultivation. A massive earthquake struck the city on February 14, 1619. It killed about 400 people and destroyed most buildings.

Trujillo played a vital role in the independence of Peru. Proclamation of independence from Spain happened in the Historic Centre on December 29, 1820. In 1822, the Congress of Peru named it the "Meritorious City and Faithful to the Fatherland."
