When the Ottoman fleet appeared in the Cyclades, she returned to Tinos to take further action. She sold her personal jewelry to pay for the equipment of 200 men. She also used these funds to care for 2,000 people who had survived the first siege of Missolonghi. Her efforts helped sustain both the fighters and the civilians caught in the conflict.

Her personal life was also marked by significant political tension. Manto became engaged to Demetrios Ypsilantis, a man of high status. However, powerful politicians like Ioannis Kolettis opposed the union. They feared the political influence of combining the Mavrogenis and Ypsilantis families. These conflicts eventually led to her exile from Nafplio. After the war ended, the leader Ioannis Kapodistrias recognized her service. He awarded her the rank of Lieutenant General and gave her a home in Nafplio. She also possessed a valuable sword with a Greek inscription, which she later gave to Kapodistrias.
