Macquarie was born on the island of Ulva in the Inner Hebrides. His parents were relatively poor and worked as sub-tenants on a small farm. His father worked as a carpenter and a miller. Despite their modest means, Macquarie received an education in Edinburgh. He studied arithmetic and English at a school there. In 1776, he joined the 84th Regiment of Foot. He traveled to North America to fight in the American War of Independence. During this time, he earned the rank of ensign.
After the war in America, Macquarie served in several other parts of the world. He spent significant time in India with the 77th Regiment of Foot. He participated in the Third Anglo-Mysore War and the Siege of Seringapatam in 1799. 

In 1809, the British government appointed Macquarie as the Governor of New South Wales. He arrived at Sydney Cove on December 31, 1809. The government chose an army commander instead of a naval officer for this role. They hoped he could manage the New South Wales Corps. This military group had previously mutinied against the former governor, William Bligh. This event was known as the Rum Rebellion. Macquarie's first task was to restore lawful government and order. 
Macquarie worked to transform the settlement into an organized urban environment. He designed the street layouts for modern central Sydney. He also planned the layouts for several settlements in Western Sydney. He ordered a new street design for Hobart in Van Diemen's Land. 

His social policies were also very influential during his eleven-year term. Macquarie favored the liberal treatment of ex-convicts, who were known as emancipists. He appointed these individuals to important government positions. He also provided them with generous land grants to help them succeed. Macquarie expressed a desire to treat Aboriginal peoples with kindness. He even established the first school specifically for Aboriginal children. 
Macquarie's era was marked by a tension between his vision and the British government. He often built expensive public structures despite a British ban on such projects. These buildings included the Parramatta Female Orphan School and St James Church. He also supported exploration of the Australian interior. In 1813, he authorized an expedition that successfully crossed the Blue Mountains. This expedition led to the establishment of Bathurst, the first inland British settlement. His combined efforts in architecture, law, and exploration left a permanent mark on the region.
