Joanna was the second daughter and third surviving child. She had several siblings, including Catherine, who became the Queen of England. Initially, Joanna was not expected to inherit the thrones of Castile or Aragon. However, a series of deaths changed her destiny. Between 1497 and 1500, her brother John, her sister Isabella, and her nephew Miguel all died. These events made Joanna the heir presumptive to both kingdoms.

To prepare for her role, Joanna received a rigorous academic and royal education. Her tutors included the Dominican priest Andrés de Miranda and the educator Beatriz Galindo. She studied canon and civil law, genealogy, and history. Joanna also mastered grammar, mathematics, philosophy, and multiple languages. She became fluent in French and Latin. In addition to academic subjects, she learned court etiquette, dancing, and music. She was skilled at playing the clavichord, the guitar, the lute, and the monochord. She also practiced outdoor pursuits like hawking and hunting.

In 1496, a political marriage was arranged to strengthen royal power.

At age 16, Joanna was betrothed to 18-year-old Philip of Austria. Philip was the son of Holy Roman Emperor Maximilian I. This marriage was a strategic alliance between the Habsburg and Trastámara families. The goal was to increase security against the growing power of France. After a proxy marriage in Valladolid, the formal ceremony occurred in Lier on 20 October 1496.

Between 1498 and 1507, Joanna and Philip had six children. These children, including Charles V, grew up to be various emperors and queens.

Political tension rose when Joanna's mother died in 1504. Joanna became the queen regnant of Castile. However, her father, Ferdinand II, did not want to relinquish control. He proclaimed himself the administrator and governor of the kingdom. In 1506, Philip became king of Castile through his marriage to Joanna. This began the rule of the Habsburgs in the Spanish kingdoms.

Conflict arose as both Ferdinand and Philip vied for authority. Ferdinand convinced the Cortes that Joanna's illness prevented her from governing. This allowed him to act as her guardian and administrator.

Tragedy struck in September 1506 when Philip died in Burgos. He died after a five-day illness, possibly from typhoid fever. Rumors even suggested that Ferdinand had poisoned him. Following Philip's death, Joanna's son Charles I became the heir. Despite being the ruling queen, Joanna was declared insane. Her father ordered her confinement in the Royal Palace in Tordesillas. She remained confined there even after she inherited the kingdom of Aragon in 1516. Although she was a co-monarch in name, she had almost no influence on national policy.

Historians continue to debate the true nature of Joanna's life and mental state. They question whether her alleged madness was real or a political tool. Some believe her family used her condition to discredit her and seize her power.
