As a young man, he worked many hard jobs. He loaded railway boxcars and worked in logging in Idaho. He also worked as a radio announcer in 1945. Eventually, he hitchhiked to Hollywood to find film work. He made his movie debut in a film called The Farmer's Daughter. At his first studio, his last name was changed from Aurness to Arness.

During World War II, Arness served in the United States Army. He wanted to be a naval fighter pilot. However, he was too tall for that job. He stood 6 feet and 7.5 inches tall. The height limit for aviators was only 6 feet and 2.5 inches. He was drafted into the Army in March 1943. As a rifleman, he landed on Anzio Beachhead in Italy. Because of his height, he was the first man sent into the water. He had to check how deep the water was. He was badly wounded in his right leg during the battle.

Before acting, Arness performed manual labor, such as loading railway boxcars and logging in Idaho. In 1945, he worked as a radio announcer at station WLOL in Minneapolis. To pursue film work, he hitchhiked to Hollywood. At RKO, the studio changed his surname from Aurness to Arness.

His military service during World War II profoundly shaped his life and health. Arness originally hoped to become a naval fighter pilot. Arness's acting career included diverse roles beyond the Western genre. While he was often identified with Westerns, he also appeared in science-fiction films. These included The Thing from Another World, where he played the titular character, and Them!. He formed a close friendship with actor John Wayne. Arness appeared in several supporting roles in films starring Wayne, such as Hondo and The Sea Chase. He also starred in the 1988 television remake of Red River, playing the role of Tom Dunson. Although an urban legend suggests Wayne offered him the role in Gunsmoke, the truth is that Wayne simply recommended him. Wayne appeared in a prologue to introduce Arness as Matt Dillon in 1955.

Gunsmoke became a massive success in television history. The series featured Arness in every one of its 20 seasons. It became the longest-running primetime drama series in American television history by its conclusion in 1975. The show aired 179 more episodes beyond its initial run. For 13 seasons, the show remained in the top 10 ratings, including four consecutive seasons at number one. Arness's co-stars, including Milburn Stone and Amanda Blake, also achieved worldwide fame. In Europe, Arness gained cult status for his role as Zeb Macahan in the series How the West Was Won.

He famously disdained publicity and even banned reporters from the Gunsmoke set. His stunt double, Ben Bates, noted that Arness possessed a joyful side, often laughing from his "toes to the top of his head." This laughter was sometimes so uncontrollable that filming on the Gunsmoke set had to be suspended. His personal life included marriage to Virginia Chapman in 1948 and later to Janet Surtees in 1978. He was the father of Rolf, Craig, and Jenny Lee Arness.

Arness received numerous accolades for his contributions to the entertainment industry. He earned a star on the Hollywood Walk of Fame at 1751 Vine Street. In 1981, he was inducted into the Western Performers Hall of Fame. His impact was recognized by major publications; People magazine ranked him sixth among the "top 25 television stars of all time" in 1989. TV Guide also placed him on its list of the "50 Greatest TV Stars of All Time" in 1996. He received three Emmy Award nominations between 1957 and 1959 for his dramatic performances. Arness died of natural causes on June 3, 2011, at the age of 88.
