Between 1723 and 1725, he served as the master of the chapel for Count Kinsky in Prague. In 1726, he established a violin school that attracted students from across Europe. Tartini's life was marked by significant personal and legal challenges. Born on April 8, 1692, in Pirano, he was the son of Gianantonio and Caterina Zangrando. While his parents intended for him to become a Franciscan friar, he instead studied law at the University of Padua. After his father died in 1710, Tartini married Elisabetta Premazore. This marriage caused conflict because of her social standing. Following legal accusations involving his wife and Cardinal Giorgio Cornaro, Tartini fled to the monastery of St. Francis in Assisi. There, he continued his musical education under Bohuslav Matěj Černohorský.

As a performer, Tartini used some of the finest instruments in history. In 1711, he began playing an Antonio Stradivarius violin known as the ex-Vogelweith. In 1715, he became the first known owner of another Stradivarius violin. He eventually passed this instrument to his student, Salvini, who later gave it to the virtuoso Karol Lipiński. This specific instrument is now famously known as the Lipinski Stradivarius. These high-quality tools were essential for his complex performances.

One of Tartini's greatest scientific contributions was his study of acoustics. He is credited with discovering sum and difference tones, which are mathematical acoustic phenomena. When playing double-stops, or two notes at once, a third tone called the "terzo suono" can be heard. This discovery was highly practical for improving musical intonation. He detailed these findings in his 1754 treatise, "Trattato di musica secondo la vera scienza dell'armonia." His work on ornamentation also influenced many other musicians, including Leopold Mozart.

Tartini's most enduring legacy is the "Devil's Trill Sonata." This solo violin piece is famous for its technically demanding double-stop trills. The piece is considered difficult even by modern performance standards. A popular legend suggests the melody came to him in a dream. In this dream, the Devil appeared at his bedside to play the violin.
