The plant then uses S-Adenosyl methionine (SAM) to methylate the caffeic acid. This step forms ferulic acid, which is then converted to feruloyl-CoA by the enzyme 4-hydroxycinnamoyl-CoA ligase (4CL). The process continues as feruloyl-CoA is reduced to coniferaldehyde by cinnamoyl-CoA reductase (CCR). Then, cinnamyl-alcohol dehydrogenase (CAD) or sinapyl-alcohol dehydrogenase (SAD) reduces that into coniferyl alcohol. This alcohol is converted into an ester called coniferyl acetate. Finally, the enzyme eugenol synthase 1 uses NADPH to transform coniferyl acetate into eugenol.

Eugenol is found in very high concentrations in certain plant parts. In clove bud oil, eugenol makes up 80% to 90% of the total composition.
