The history of European contact began in 1606. A Spanish expedition led by Pedro Fernandes de Queirós arrived at Big Bay. This bay is on the north side of the island. Queirós named the land La Estensen. He believed he had found the Great Southern Continent, known as Terra Australis. The explorers stayed at an anchorage called Santiago for one month. During this time, armed sailors went inland to find provisions. A conflict occurred with a local chief, which resulted in the chief's death. Queirós intended to found a city called Nova Jerusalem. However, the expedition faced difficulties. The lead ship became separated from the others. Luis Váez de Torres, the second-in-command, searched for the ship but could not find it. Torres eventually sailed west and discovered the strait that now bears his name.

Following these early visits, Europeans did not return for 160 years. Louis de Bougainville visited in 1768, and James Cook arrived in 1774. During the 19th century, settlers from Australia, Britain, France, and Germany arrived. In 1878, the United Kingdom and France declared the New Hebrides to be neutral territory. This led to the creation of the Anglo-French Joint Naval Commission in 1887. In 1889, a group called Franceville attempted to become an independent commune. It was led by Ferdinand-Albert Chevillard. Franceville was notable for practicing universal suffrage. This meant people could vote regardless of their sex or race. This experiment was short-lived. The Naval Commission suppressed the commune by June 1890. In 1906, the administration became a more structured British-French Condominium.

Geological studies also shaped the understanding of the island. Between 1903 and 1905, Australian geologist Sir Douglas Mawson conducted major studies. He explored the islands with W.T. Quaife. They traveled aboard the ship Ysabel. Mawson produced detailed geological maps of Efate and Santo. His report, "The Geology of the New Hebrides," was published in 1905. This work was his first major independent geological study. During World War II, the island's role changed significantly. Following the attack on Pearl Harbor, the United States used Santo as a military base. It served as a naval harbor and an airfield.

The Seabees of the 40th Naval Construction Battalion built Luganville Airfield, also called Bomber Field #3, in 1943. Other airfields included Bomber Field No. 2 and Palikulo Bay Airfield. The SS President Coolidge, a converted luxury liner, was sunk by a sea mine during the war. This shipwreck is now a popular diving site.

The island experienced political tension during the transition to independence. In 1980, a rebellion known as the "Coconut War" occurred. This happened as power transferred from the colonial Condominium to the new nation of Vanuatu. The Nagriamel movement, led by Jimmy Stevens, declared Espiritu Santo independent. They proclaimed the Republic of Vemerana on May 28, 1980. The movement was backed by the Phoenix Foundation and some American libertarians. They hoped to create a tax-free haven. While France recognized the independence of Vemerana, the new government of Vanuatu did not. Eventually, the army of Papua New Guinea was requested to help. They suppressed the rebellion and kept Espiritu Santo as part of Vanuatu.

Today, the economy relies on several different sectors. Many people engage in subsistence farming and fishing. Others participate in the tourist trade. Cash-crop farming is also important. Farmers grow cocoa beans, kava, and peanuts. Copra is another major crop.
