His freshman professors included the famous mathematician John Forbes Nash Jr. He earned both his bachelor's and master's degrees in electrical engineering in 1962. During this time, he completed internships at Bell Labs in 1960 and 1962. His supervisor at the lab was John Larry Kelly Jr. He finished his Ph.D. in electrical engineering in 1964. His doctoral advisors included Claude Shannon and Robert G. Gallager. Other advisors were Peter Elias and John Wozencraft.

Berlekamp held several important professional roles throughout his life. He taught electrical engineering at UC Berkeley from 1964 to 1966. Then, he worked as a mathematics researcher at Bell Labs. He returned to Berkeley in 1971 as a professor of mathematics and computer science. In that role, he advised more than twenty doctoral students. He also led Cyclotomics, Inc. in the mid-1980s. This was a corporation that developed error-correcting code technology. His professional excellence earned him many prestigious memberships and awards.

His contributions were recognized by many scientific organizations. He was a member of the National Academy of Engineering in 1977. In 1999, he joined the National Academy of Sciences. He became a Fellow of the American Academy of Arts and Sciences in 1996. He also became a fellow of the American Mathematical Society in 2012. Berlekamp received the IEEE Richard W. Beyond formal research, Berlekamp loved recreational mathematics. He was a close friend of the columnist Martin Gardner. He helped found Gathering 4 Gardner, an organization celebrating Gardner's legacy. This group holds conferences every two years. Berlekamp served on the board of directors for many years. He lived in Piedmont, California, for much of his life. He passed away in April 2019 at the age of 78. His death was caused by complications from pulmonary fibrosis.
