The virus works by entering a person's cells to make copies of itself. It has a very large genome, which is the set of instructions for the virus. This large genome helps it fight against a person's natural defenses. The virus can even make its own machinery to copy its DNA. Once it is inside a cell, the virus uncoats and begins to replicate. This replication happens in the cytoplasm, which is a part of the cell. Finally, the new viruses are assembled and released to find new cells.

People have studied this virus for a very long time. An English farmer named Benjamin Jesty used cowpox to protect his family in 1774. He was not a scientist, so he did not publish his work. Later, in 1796, a doctor named Edward Jenner tested the idea more carefully. He took fluid from a cowpox sore on a woman named Sarah Nelmes. He used that fluid to help a boy named James Phipps. Jenner proved that this could protect people from the deadly smallpox virus.

Historically, people often caught the virus from infected cows. Today, human cases are very rare and often come from domestic cats. These cats usually catch the virus from woodland rodents, specifically voles. When humans are infected, they often develop painful, localized pustular lesions. These blisters usually appear on the hands or face. The incubation period, or the time between infection and symptoms, is about 9 to 10 days. The virus is most prevalent during the late summer and autumn months.

Inside the cell, the cowpox virus follows a specific cycle to multiply. The virus is a double-stranded DNA virus with an envelope surrounding the virion. The CPXV genome is very large, measuring over 220k base pairs. This makes it the largest genome in the Orthopoxvirus species. This large genome allows the virus to encode its own transcription and DNA replication machinery. Once the virus enters a host cell, the virion uncoats. The replication process then takes place within the cytoplasm, which is the fluid inside the cell. After the virus copies itself, the new virions are assembled and released from the host cell to find new targets.

The structure of the CPXV genome is quite complex and divided into specific parts. It has two end regions called R1 and R2. There is also a main central region that is about half the size of the entire genome. The genome contains inverted terminal repeats at the terminal sites. These repeats measure around 10k base pairs and can be repeated up to thirty times. Interestingly, the CPXV genome only encodes about 30% to 40% of products involved in pathogenesis, or the process of causing disease. This unique genetic makeup makes the virus very capable of mutating into different strains.

One of the most remarkable features of cowpox is how it fights the immune system. The genome is arranged so that the ends contain genes used to evade host defenses. The virus can block the secretion of cytokines and chemokines, which are signaling proteins used by the immune system.

Because of its large genome, CPXV has more cytokine responses to fight back than other poxviruses. It encodes various receptors, such as TNF, CrmB, CrmC, CrmD, and CrmE proteins. It also encodes lymphotoxins like IL-1ß and IFN-y. These proteins play a crucial role in how the virus interacts with the body's natural defenses.

The history of cowpox is tied to one of the greatest medical discoveries in human history. In 1774, an English farmer named Benjamin Jesty used cowpox to immunize his family. He transferred pieces of infected cow udder into their skin to protect them from smallpox. However, Jesty was not a scientist and did not publish his findings. Later, in 1796, Edward Jenner tested this theory more formally. He took fluid from a cowpox lesion on a milkmaid named Sarah Nelmes. He applied this fluid to an eight-year-old boy named James Phipps. After Phipps recovered from the mild cowpox, Jenner exposed him to smallpox. Phipps did not get sick, proving that cowpox provided immunity against the deadly Variola virus.
