He is a former professional mixed martial artist and a 5th degree black belt in Brazilian Jiu-Jitsu (BJJ). Penn achieved a rare feat by winning UFC titles in two different weight classes. He was the second fighter in the history of the Ultimate Fighting Championship (UFC) to do this. Before his career in mixed martial arts, he was a world-class grappler. He was the first non-Brazilian to win the World Jiu-Jitsu Championship at the black belt level. His skill and impact earned him induction into the UFC Hall of Fame in 2015.

Penn's journey began with a dedicated study of Brazilian Jiu-Jitsu. At age 17, he started training after meeting his neighbor, Tom Callos. Penn later moved to San Jose, California, to train at the Ralph Gracie BJJ academy. He progressed through the ranking system by earning a purple belt and then a brown belt. In 2000, he was awarded his black belt by André Pederneiras at Nova União. This achievement followed a series of successful tournament placements. His success in grappling led to a career in the Ultimate Fighting Championship. Penn made his debut at UFC 31 on May 24, 2001. He quickly proved he was more than just a grappler by using striking skills. He earned knockout victories over fighters like Din Thomas and Caol Uno. One of his most famous moments occurred in 2004 at UFC 46. He moved up in weight to challenge the Welterweight Champion, Matt Hughes. Despite being the underdog, Penn won using a rear naked choke. This move involved controlling the opponent's back and applying pressure to the neck. This victory remains one of the greatest upsets in mixed martial arts history.

Penn's career involved many different weight divisions and organizations. He competed in the Featherweight, Lightweight, Welterweight, Middleweight, and Heavyweight divisions. During a period away from the UFC, he signed with the Japanese promotion FEG. While fighting for FEG, he won a middleweight bout against Rodrigo Gracie by decision. He also defeated Duane Ludwig using an arm triangle choke, which is a submission using the arms to compress the neck. However, the UFC stripped him of his welterweight title during this time. They claimed he had breached his contract by signing with another organization. Penn later filed a lawsuit, claiming his UFC contract had already expired.

In 2008, Penn returned to the UFC to claim the Lightweight Championship. He fought Joe Stevenson at UFC 80 on January 19, 2008. Penn used an uppercut to knock Stevenson down early in the first round. He later won the fight in the second round with a rear naked choke. This victory made him only the second person, after Randy Couture, to hold UFC titles in two weight classes.
