Most of the time, these sori are located on just one side of a leaf vein. In rare cases, they may appear in pairs on a single vein, but they will not curve over that vein. A thin, membranous flap called an indusium arises along one edge of the sorus to protect it.

Additionally, the leaf stalks, or petioles, have a specific internal structure. They contain two vascular bundles that unite to form an X-shape when viewed in a cross-section near the tip. The stalks of the sporangia, which hold the spores, are only one cell wide in the middle.

The way scientists classify Aspleniaceae has changed significantly over time. Edward Newman first described the family in 1840. At that time, he included three different genera: Athyrium, Asplenium, and Scolopendrium. Modern science has since reorganized these groups. Athyrium is now placed in a separate family called Athyriaceae because it is not strongly related to spleenworts. Scolopendrium is now considered a synonym of Asplenium. This means scientists now view them as the same group rather than two different ones.

Recent classification efforts have focused on using molecular phylogenetic studies to group plants. These studies look at the genetic relationships between species. In 2016, the Pteridophyte Phylogeny Group (PPG I) adopted a narrow circumscription for the family. This narrow view, known as Aspleniaceae s.s., recognizes only two genera: Asplenium and Hymenasplenium. Previously, the genus Asplenium had been split into about a dozen different genera, such as Diella, which is found only in Hawaii. However, consensus shows these are actually nested within the larger Asplenium group.

There are different ways to define the boundaries of this family. The PPG I system places the family in the suborder Aspleniineae within the order Polypodiales. This group is part of a larger clade known as eupolypods II. Some researchers, such as Christenhusz and Chase, proposed a much broader circumscription. In their view, the family included all the separate families that PPG I places in the suborder Aspleniineae. Under this broader view, those families are reduced to subfamilies. The Plants of the World Online database uses this broader classification, which lists 24 different genera.

The two main groups in the narrow classification show very different scales of diversity. The genus Asplenium is the largest, containing about 700 different species. These species are found almost everywhere in the world, except for Antarctica and some high Arctic regions. The second genus, Hymenasplenium, is much smaller with about 40 species. These are found specifically in tropical and subtropical areas. This wide distribution makes the Aspleniaceae a very important group for studying global plant life.
