After attending high school in the capital city of Luanda, Neto worked in colonial health services. He later moved to Portugal to study medicine at the University of Coimbra and the University of Lisbon. During his time as a medical student, he engaged in covert revolutionary activities against the Portuguese government. This activism led to several arrests by the PIDE, the security police of the Estado Novo regime. He was arrested in 1951, 1952, and again in 1955, being held until 1957. After finishing his studies and marrying Maria Eugénia da Silva, he returned to Angola in 1959. However, he was arrested a fourth time in 1960, after which he escaped prison to lead the armed struggle for independence.

The political organization known as the MPLA was formed in December 1956. It resulted from a merger between the Angolan Communist Party (PCA) and the Party of the United Struggle for Africans in Angola (PLUAA). Viriato da Cruz served as the first Secretary General, while Neto became the president.

During the struggle for independence, Neto sought international support for his movement. In 1962, he visited Washington, D.C., to ask the Kennedy administration for aid. The United States declined, choosing instead to support the National Liberation Front of Angola (FNLA) due to oil interests. Seeking other allies, Neto met Che Guevara in 1965 and established strong ties with Cuba. He also conducted official diplomatic trips to nations like Romania, Bulgaria, and Yugoslavia to discuss African political matters.

Following the Carnation Revolution in Portugal in April 1974, the path to independence became clear. Three different political factions competed for control of the future Angolan state. On 11 November 1975, Angola officially gained independence from Portugal. The MPLA seized control of the capital, Luanda, and Neto became the nation's leader.

He established a one-party state based on the official doctrine of Marxism-Leninism. His government maintained close relationships with the Soviet Union and other Eastern Bloc nations. This era also saw the beginning of the Angolan Civil War, which started in 1975 and continued until 2002. During this conflict, the MPLA received significant military assistance from Cuba to fight against the FNLA, UNITA, and South African forces.

Neto's presidency faced significant internal challenges, including a period of violent political unrest. In 1977, a movement known as Fractionism attempted a coup d'état. This group was inspired by the Organização dos Comunistas de Angola. In the aftermath of this attempt, tens of thousands of people associated with the movement were executed over a two-year period. While Neto only personally ratified the death sentence of the leader Nito Alves, the period was marked by intense repression. In response to the "excesses" committed by the Directorate of Information and Security, Neto eventually decided to dissolve that specific organization.

Beyond his political life, Neto was a prolific and celebrated writer. He published three books of poetry during his lifetime, including the collection *Sacred Hope*, published in 1974.

His poetic works were mostly written between 1946 and 1960 while he was living in Portugal. Many of his poems were so influential that they became national anthems. He was the first member of the Anglo Writers Union and the Center for African Studies in Lisbon. His literary talent earned him the Lotus Prize from the Conference of Afro-Asian Writers. He is widely considered to be Angola's most important poet.

Neto's health declined due to pancreatic cancer and chronic hepatitis. He traveled to the Soviet Union several times to receive treatment from highly skilled medical professionals. He died in Moscow on 10 September 1979, just one week before his 57th birthday.
